Uses of Letrozole Tablets
Letrozole Tablets (Letromina) is used to treat breast
cancer in post-menopausal women that is, women
who no longer have periods, either naturally due to
their age or after surgery or chemotherapy.
Letrozole tablets are used for adjuvant treatment of
postmenopausal women with hormone receptor
positive invasive early breast cancer. It is also used
to treat invasive early breast cancer in post
menopausal women who have received prior
treatment with a medication called Tamoxifen for
five years.

Uses of Letrozole Tablets
Letrozole is also used as a first-line treatment in
postmenopausal women with advanced breast cancer and
advanced breast cancer after relapse or disease
progression, in women with natural or artificially induced
postmenopausal endocrine status, who have previously
been treated with anti-oestrogens.
Letrozole works by decreasing the amount of estrogen
produced by the body. Reduction of estrogen can slow or
stop the growth of some types of breast cancer cells that
require estrogen to grow.
Off label uses include ovarian stimulation by fertility
doctors because it has fewer side-effects than
clomiphene, treatment for gynecomastia, and treatment of
endometriosis.

Recommended Adult Dosage
The recommended dose of Letrozole Tablets
(Letromina) is one 2.5 mg tablet administered once
a day, with or without meals. The recommended
adult dosage of is given below:
➔Use in Adjuvant Treatment and Extended
Adjuvant Treatment of Early Breast Cancer: In
the adjuvant and extended adjuvant setting, the
treatment with Letrozole should continue for 5
years or until disease relapse/recurrence occurs,
whichever comes first.
➔Use in First and Second-Line Treatment of
Advanced Breast Cancer: In patients with
advanced disease, treatment with Letrozole should
continue until tumor progression is evident.

Contraindications
Letrozole tablets (Letromina 2.5mg) are
contraindicated in patients with the following
conditions:
➔Known hypersensitivity to Letrozole or any
inactive ingredient of this medication.
➔Use of Letrozole is contraindicated in women of
premenopausal endocrine status, including
pregnant women.

Side Effects of Letrozole Tablets
The most common possible side effects of
Letrozole tablets (Letromina 2.5 mg)
occuring in 20% or more of the patients
undergoing treatment were hot flashes,
arthralgia (joint pain), flushing, asthenia,
edema, arthralgia, headache, dizziness,
hypercholesterolemia, increased sweating,
bone pain and musculoskeletal problems.

Warnings And Precautions
➔Use of Letrozole Tablets (Letromina) may cause
decreases in bone mineral density (BMD). Bone mineral
density monitoring is advised.
➔Caution is advised when driving or using machinery as
fatigue, dizziness, and somnolence have been reported
with the use of Letrozole tablets.
➔Subjects with cirrhosis and severe hepatic impairment
who were dosed with 2.5 mg of Letrozole tablets
experienced approximately twice the exposure to
Letrozole as healthy volunteers with normal liver function.

Warnings And Precautions
➔The potential risk/benefit to patients with creatinine
clearance less than 10 mL/min should be carefully
considered before administration of Letrozole tablets.
➔In patients whose menopausal status is unclear,
luteinising hormone (LH), follicle-stimulating hormone
(FSH) and/or oestradiol levels should be measured before
initiating treatment with Letrozole tablets.
➔Co-administration of Letromina (Letrozole tablets) with
Tamoxifen, other anti-oestrogens or oestrogen-containing
therapies should be avoided as these substances may
diminish the pharmacological action of Letrozole.

11. Letrozole Tablets
Overdose Symptoms & Treatment
As there have been very few isolated cases of
Letrozole overdose that were reported, it is not clear
what exactly to expect from an overdose of
Letrozole tablets. In most cases, treatment for a
overdose of Letrozole tablets will not be necessary.
The single highest reported dose taken was 62.5 mg
or 25 tablets. No serious side effects were reported.
No specific treatment for Letrozole overdose is
recommended, although emesis could be induced if
the patient is alert. Supportive care with frequent
monitoring of vital signs is recommended.
