Abstract: The human intestinal tract harbours a complex microbial ecosystem which plays a key role in nutrition and health. Interactions between food constituents, microbes and the host organism derive from a long co-evolution that resulted in a mutualistic association. Current investigations into the human faecal metagenome are delivering an extensive gene repertoire representative of functional potentials of the human intestinal microbiota. The most redundant genomic traits of the human intestinal microbiota are identified and thereby its functional balance. These observation point towards the existence of enterotypes, i.e. microbiota sharing specific traits but yet independent of geographic origin, age, sex etc.. It also shows a unique segregation of the human population into individuals with low versus high gene-counts. In the end, it not only gives an unprecedented view of the intestinal microbiota, but it also significantly expands our ability to look for specificities of the microbiota associated with human diseases and to ultimately validate microbial signatures of prognostic and diagnostic value in immune mediated diseases. Metagenomics of the human intestinal tract was applied to specifically compare obese versus lean individuals as well as to explore the dynamic changes associated with a severe calory-restricted diet. Microbiota structure differs with body-mass index and a limited set of marker species may be used as diagnostic model with a >85% predictive value. Among obese subjects; the overall phenotypic characteristics are worse in individuals with low gene counts microbiota, including a worse evolution of morphometric parameters over a period of 10 years, a low grade inflammatory context also associated with insulin-resistance, and the worst response to dietary constraints in terms of weight loss or improvement of biological and inflammatory characteristics. Low gene count microbiota is also associated with less favourable conditions in inflammatory bowel disease, such as higher relapse rate in ulcerative colitis patients. Finally, microbiota transplantation has seen a regain of interest with applications expanding from Clostridium difficile infections to immune mediated and metabolic diseases. The human intestinal microbiota should hence be regarded as a true organ, amenable to rationally designed modulation for human health.

Commentary Origins and evolution of the Western diet: health implications for the 21st century1,2 Loren Cordain, S Boyd Eaton, Anthony Sebastian, Neil Mann, Staffan Lindeberg, Bruce A Watkins, James H O’Keefe, and Janette Brand-Miller ABSTRACT There is growing awareness that the profound changes in the envi- ronment (eg, in diet and other lifestyle conditions) that began with the introduction of agriculture and animal husbandry �10 000 y ago occurred too recently on an evolutionary time scale for the human genome to adjust. In conjunction with this discordance between our ancient, genetically determined biology and the nutritional, cultural, and activity patterns of contemporary Western populations, many of the so-called diseases of civilization have emerged. In particular, food staples and food-processing procedures introduced during the Neolithic and Industrial Periods have fundamentally altered 7 cru- cial nutritional characteristics of ancestral hominin diets: 1) glyce- mic load, 2) fatty acid composition, 3) macronutrient composition, 4) micronutrient density, 5) acid-base balance, 6) sodium-potassium ratio, and 7) fiber content. The evolutionary collision of our ancient genome with the nutritional qualities of recently introduced foods may underlie many of the chronic diseases of Western civilization. Am J Clin Nutr 2005;81:341–54. KEY WORDS Westernized diets, chronic disease, processed foods, genetic discordance, hunter-gatherers, human evolution EVOLUTIONARY DISCORDANCE Evolution acting through natural selection represents an on- going interaction between a species’ genome and its environment over the course of multiple generations. Genetic traits may be positively or negatively selected relative to their concordance or discordance with environmental selective pressures (1). When the environment remains relatively constant, stabilizing selec- tion tends to maintain genetic traits that represent the optimal average for a population (2). When environmental conditions permanently change, evolutionary discordance arises between a species’ genome and its environment, and stabilizing selection is replaced by directional selection, moving the average population genome to a new set point (1, 2). Initially, when permanent environmental changes occur in a population, individuals bear- ing the previous average status quo genome experience evolu- tionary discordance (2, 3). In the affected genotype, this evolu- tionary discordance manifests itself phenotypically as disease, increased morbidity and mortality, and reduced reproductive success (1–3). Similar to all species, contemporary humans are genetically adapted to the environment of their ancestors—that is, to the environment that their ancestors survived in and that conse- quently conditioned their genetic makeup (1–3). What scientific mechanism for evolution did Charles Darwin and Alfred Wallace propose in 1858? Describe how this concept works. What is the difference between microevolution and macroevolution? Solution 1. Charles Darwin during 1858 had proposed the Theory of Evolution by Natural Selection, where he has said that organisms change over time as a result of changes in heritable physical or behavioral traits. Changes that allow an organism to better adapt to its environment will help it survive and have more offspring. Alfred Wallace who is also called the Father of Biogeography, studied the geographical distribution of animal species and said the evolution is caused due to environmental pressures on varieties of species it forces different species to adapt themselves to the local conditions, leading to different populations in different locations. Both groups of scientists have said that natural selection is the single most and important factor in evolutionary changes seen in species. Micro Evolution is caused due to Mutation, Migration or Gene Flow, Genetic Drift and Selection (natural and artificial). Macro Evolution: It refers to major evolutionary changes over time, the origin of new types of organisms from previously existing, but different, ancestral types.It is an evolution on a larger scale where we find the descent of many species from one common ancestor over billions of years.It is very difficult to see the Macro Evolution happening because it takes a longer time to happen. This keynote will begin with a personal case study exploring how the speaker became the target of a generative AI scam, highlighting the convincing nature of these deceptions. Building upon this experience, the talk will delve into the broader implications of generative AI for the spread of misinformation and the erosion of trust in online spaces. As the line between truth and falsehood becomes increasingly blurred, the role of education in equipping individuals with the critical thinking skills needed to navigate this complex landscape becomes paramount. The keynote will argue that empowering postgraduate social science researchers with the tools to critically engage with generative AI is not only essential for their own work but also for the wider fight against misinformation.
