I.INTRODUCTION
STANDARDIZATION OF
PLECTRANTHUS AMBOINICUS
Plectranthus amboinicus Lour. known as country borage, Indian
borage in English and Patta ajwain , Patharcur in Hindi. Leaves roots
and stems are used as carminative, digestive, stomachic,
anthelmintic, espectorant, diuretic, otalgia, anorexia, diarrhea
and cholera especially in children, convulsions epilepsy,
chronic asthma, bronchitis, renal and vesical calculi and
malarial fever . Leaf consists of oleanolic acid, betulin and other
triterpenoids. The plant yields barbatusol and β sitosterol.
Plectranthus amboinicus
Plectranthus amboinicus Lour. (Lamiaceae) has got high reputation in
traditional medicinal practice for its remarkable medicinal properties. The
entire plant, especially the leaves are recognized as valuable drugs
and frequently used by many ancient physicians.

Many pharmacological properties such as Urolithiasis, Antiepileptic,
Antitumorogenic, Antimutagenic, Radioprotective, Neuropharmacological,
and Antimicrobial have been reported for the leaves of P. amboinicus .
The leaves exhibit significant antioxidant potency, reducing power, superoxide
anion radical scavenging ability, nitric oxide radical scavenging, and ferrous
ion chelating ability.
They contain essential oils,flavonoids, and terpenes which possess inhibitory
effect against Gram-positive and Gram-negative bacteria .
Antimutagenic, Antitumorogenic and Antigenotoxic effects of Indian borage
leaves have also been documented.
The study included Antioxidant, Antiplatelet, Antibacterial, and Anticancer
properties of stem that could have the potential as neutraceuticals (any product
derived from food sources with extra health benefits in addition to the basic
nutritional value found in foods) and functional food ingredients.
II. MATERIALS AND
METHODS
PREPARATION OF EXTRACT
 The plant
Plectranthus
amboinicus Lour,
was collected .
 The plant was
identified and
authenticated.
 The leaves of
Plectranthus
amboinicus (Lour)
were shade dried at
room temperature.

 Then the shade dried leaves were powdered to get a coarse
powder.
 60g of coarse powder was defatted with petroleum ether and
extracted exhaustively with 95% methanol at Temperature 60
C, in a soxhlet extractor.
 The extract was concentrated in a rotary flash evaporator.
 The residue was dried in a desiccator over sodium sulfite.
 This procedure was repeated for 5-6 times to receive sufficient
quantity of methanolic extract.
III.PHYTO CHEMICAL
INVESTIGATION
PHYTO CHEMICAL INVESTIGATION
Methanolic extract of Plectranthus
amboinicus (Lour) Spreng leaves were
subjected to further
preliminary,qualitative,phytochemical
investigation.
Qualitative Phytochemical analysis
 Chemical tests were carried out on aqueous extracts of dry leaves using
standard procedures to identify bioactive molecules.
Isolation of volatile chemical compounds
 The plant leaves were boiled for 30 minutes and the water cooled. The plant
volatiles were collected from the warm aqueous extract by head space solid
phase microextraction (HS-SPME) using a
Polydimethylsiloxae/divinylbenzene 65μm- fiber (PDMS/DBV) for a
sample/headspace equilibration time of 1 hour.
 The extracted materials were subsequently desorbed and analyzed in the
GC-MS apparatus.

GAS CHROMATOGRAPHY MASS
SPECTROMETRY
 The volatile compounds extracted by SPME were analyzed by a GC-MS electron impact
ionization (EI) method on a Varian GC equipped with an a DB-5 wax ( J&W Scientific;
column 30 m long × 0.25 mm i.d; 0.25 μm film thickness).
 Oven temperature was programmed from 400C (1 min) with an increase of 30C/min to
2350 for 9 min.
 Injections were performed with injector temperature of 230 0C, ion source temperature
2000C.
 Volume injected was 1μl of the oil.
 Helium was used as a carrier gas at a flow rate of 1ml/min.
 Mass spectra were taken at 70eV, a scan interval of 1sec and a scan range of 30-
400m/z, full scan mode that revealed the total ion current (TIC) chromatograms.
 A Finnigan SSQ 7000 MS was used to identify and quantify the individual compounds.
 The bioactive compounds were identified by calculating retention indices using a
homologous series of n-alkanes (C5-C26) and comparing retention indices (RIs) with
literature values measured on columns with identical polarities.
 The components were also identified by matching their Mass spectra Chemical
composition and Toxicological evaluation with those in the available reference.
PREPARATION OF TEST ANIMALS
 Disease free mice (n=30) and Wistar rats (n=30) of both sexes were
obtained.
 The animals were kept in cages for one week to acclimatize and
were maintained on standard animal diet and water in excess.
 However food was withdrawn 18 hours before the start of the
experiment in acute toxicity.
ACUTE TOXICITY STUDY
 The toxicity study as carried out using female and male albino
mice (20-35 g).
 The acute toxicity studies were conducted asper the OECD
guidelines 420(OECD 2000) where the limit testdose of 2000
mg/kg was used.
 The animals were divided into one control group and one
treated group, each group consisting of ten animals (5 males
and 5 females).
 Observations were made at 2,4,8hrs for seven days for
bodyweight, treatment related changes likerespiration rate
and heart rate and behavioral signs like apathy,reduced
locomotor behavior.
SUB ACUTE-TOXICITY STUDY
Healthy adult Female albino mice weighing 20-30 gm
were divided in to 3 groups of 6 animals each and were
housed under standard conditions and room
temperature (25±2oC).
The control animals(Group-I) received 0.5ml of vehicle
alone and the other two groups(Group-II &III) have
received PA extract for 28 days at doses of 200,400
mg/Kg body wt respectively.
Toxic manifestations and mortality were monitored
daily and body wt. changes were recorded every 7
days till the end of the study.
IV. RESULT/CONCLUSION
STANDARDIZATION OF PLANT
MATERIAL
To ensure reproducible quality of herbal products, proper control of
starting material is utmost essential.
According to WHO, the macroscopic and microscopic
description of a medicinal plant is the first step towards
establishing its identity and purity and should be carried out before
any tests are undertaken.
The test for loss on drying determines both water and volatile
matter.
This is especially for materials that absorb moisture easily or
deteriorate quickly in presence of water.
The residue remaining after incineration of plant material is the
ash content, which represents inorganic salts naturally occurring in
crude drug.
Another concern related to safety of herbals is presence of
contaminants such as heavy metals. The plant material was
subjected to heavy metal Analysis.
ACUTE TOXICITY STUDY
 The acute toxicity study was conducted as per the OECD guidelines 420,
where the lim No test substance related mortality was observed at
2000mg/Kg and through out the observation period there were no significant
changes in the body weight and treatment related change like respiration rate
and heart rate.
 Persistent treatment related changes were observed in behavioural signs viz
apathy, reduced locomotor behavior but regained after 24 hrs. Consequently,
2000mg/Kg of plant extract found safe with less toxic effect.it test dose of
2000mg/Kg was used.
SUB ACUTE TOXICITY STUDY
The methanolic extract of Plectranthus amboinicus (Lour) Spreng at dose of
200,400 mg/kg orally for every 24 hr for 28 days did not produce any mortality in
tested animals.
No sign of observable toxicity was detected during the experimental period.
All the tested haematological parameters such as hemoglobin, R.B.C, Platelet
count, Reticulocyte count, Mean corpuscular volume, mean corpuscular
hemoglobin concentration, Percent of Neutrophils, Lymphocytes and Monocytes,
Packed cell volume and mean corpuscular hemoglobin were within normal.
W.B.C count was slightly increased in group-II and decreased in Group-III than
the Group-I.
Biochemical parameters such as serum bilirubin, Serum glutamic oxaloacetic
Transaminase, Serum Glutamic pyruvic Transaminase, Serum alkaline
phosphatase, Serum total proteins, serum total albumin, serum total globulin and
serum cholesterol and serum triglyceride were within the normal.

Standardization of plectranthus amboinicus

  • 3. STANDARDIZATION OF PLECTRANTHUS AMBOINICUS Plectranthus amboinicus Lour. known as country borage, Indian borage in English and Patta ajwain , Patharcur in Hindi. Leaves roots and stems are used as carminative, digestive, stomachic, anthelmintic, espectorant, diuretic, otalgia, anorexia, diarrhea and cholera especially in children, convulsions epilepsy, chronic asthma, bronchitis, renal and vesical calculi and malarial fever . Leaf consists of oleanolic acid, betulin and other triterpenoids. The plant yields barbatusol and β sitosterol. Plectranthus amboinicus Plectranthus amboinicus Lour. (Lamiaceae) has got high reputation in traditional medicinal practice for its remarkable medicinal properties. The entire plant, especially the leaves are recognized as valuable drugs and frequently used by many ancient physicians.
  • 4. CONT’D……….. Many pharmacological properties such as Urolithiasis, Antiepileptic, Antitumorogenic, Antimutagenic, Radioprotective, Neuropharmacological, and Antimicrobial have been reported for the leaves of P. amboinicus . The leaves exhibit significant antioxidant potency, reducing power, superoxide anion radical scavenging ability, nitric oxide radical scavenging, and ferrous ion chelating ability. They contain essential oils,flavonoids, and terpenes which possess inhibitory effect against Gram-positive and Gram-negative bacteria . Antimutagenic, Antitumorogenic and Antigenotoxic effects of Indian borage leaves have also been documented. The study included Antioxidant, Antiplatelet, Antibacterial, and Anticancer properties of stem that could have the potential as neutraceuticals (any product derived from food sources with extra health benefits in addition to the basic nutritional value found in foods) and functional food ingredients.
  • 6. PREPARATION OF EXTRACT  The plant Plectranthus amboinicus Lour, was collected .  The plant was identified and authenticated.  The leaves of Plectranthus amboinicus (Lour) were shade dried at room temperature.
  • 7. CONT’D….  Then the shade dried leaves were powdered to get a coarse powder.  60g of coarse powder was defatted with petroleum ether and extracted exhaustively with 95% methanol at Temperature 60 C, in a soxhlet extractor.  The extract was concentrated in a rotary flash evaporator.  The residue was dried in a desiccator over sodium sulfite.  This procedure was repeated for 5-6 times to receive sufficient quantity of methanolic extract.
  • 9. PHYTO CHEMICAL INVESTIGATION Methanolic extract of Plectranthus amboinicus (Lour) Spreng leaves were subjected to further preliminary,qualitative,phytochemical investigation.
  • 10. Qualitative Phytochemical analysis  Chemical tests were carried out on aqueous extracts of dry leaves using standard procedures to identify bioactive molecules. Isolation of volatile chemical compounds  The plant leaves were boiled for 30 minutes and the water cooled. The plant volatiles were collected from the warm aqueous extract by head space solid phase microextraction (HS-SPME) using a Polydimethylsiloxae/divinylbenzene 65μm- fiber (PDMS/DBV) for a sample/headspace equilibration time of 1 hour.  The extracted materials were subsequently desorbed and analyzed in the GC-MS apparatus.

  • 12. GAS CHROMATOGRAPHY MASS SPECTROMETRY  The volatile compounds extracted by SPME were analyzed by a GC-MS electron impact ionization (EI) method on a Varian GC equipped with an a DB-5 wax ( J&W Scientific; column 30 m long × 0.25 mm i.d; 0.25 μm film thickness).  Oven temperature was programmed from 400C (1 min) with an increase of 30C/min to 2350 for 9 min.  Injections were performed with injector temperature of 230 0C, ion source temperature 2000C.  Volume injected was 1μl of the oil.  Helium was used as a carrier gas at a flow rate of 1ml/min.  Mass spectra were taken at 70eV, a scan interval of 1sec and a scan range of 30- 400m/z, full scan mode that revealed the total ion current (TIC) chromatograms.  A Finnigan SSQ 7000 MS was used to identify and quantify the individual compounds.  The bioactive compounds were identified by calculating retention indices using a homologous series of n-alkanes (C5-C26) and comparing retention indices (RIs) with literature values measured on columns with identical polarities.  The components were also identified by matching their Mass spectra Chemical composition and Toxicological evaluation with those in the available reference.
  • 13. PREPARATION OF TEST ANIMALS  Disease free mice (n=30) and Wistar rats (n=30) of both sexes were obtained.  The animals were kept in cages for one week to acclimatize and were maintained on standard animal diet and water in excess.  However food was withdrawn 18 hours before the start of the experiment in acute toxicity.
  • 14. ACUTE TOXICITY STUDY  The toxicity study as carried out using female and male albino mice (20-35 g).  The acute toxicity studies were conducted asper the OECD guidelines 420(OECD 2000) where the limit testdose of 2000 mg/kg was used.  The animals were divided into one control group and one treated group, each group consisting of ten animals (5 males and 5 females).  Observations were made at 2,4,8hrs for seven days for bodyweight, treatment related changes likerespiration rate and heart rate and behavioral signs like apathy,reduced locomotor behavior.
  • 15. SUB ACUTE-TOXICITY STUDY Healthy adult Female albino mice weighing 20-30 gm were divided in to 3 groups of 6 animals each and were housed under standard conditions and room temperature (25±2oC). The control animals(Group-I) received 0.5ml of vehicle alone and the other two groups(Group-II &III) have received PA extract for 28 days at doses of 200,400 mg/Kg body wt respectively. Toxic manifestations and mortality were monitored daily and body wt. changes were recorded every 7 days till the end of the study.
  • 17. STANDARDIZATION OF PLANT MATERIAL To ensure reproducible quality of herbal products, proper control of starting material is utmost essential. According to WHO, the macroscopic and microscopic description of a medicinal plant is the first step towards establishing its identity and purity and should be carried out before any tests are undertaken. The test for loss on drying determines both water and volatile matter. This is especially for materials that absorb moisture easily or deteriorate quickly in presence of water. The residue remaining after incineration of plant material is the ash content, which represents inorganic salts naturally occurring in crude drug. Another concern related to safety of herbals is presence of contaminants such as heavy metals. The plant material was subjected to heavy metal Analysis.
  • 18. ACUTE TOXICITY STUDY  The acute toxicity study was conducted as per the OECD guidelines 420, where the lim No test substance related mortality was observed at 2000mg/Kg and through out the observation period there were no significant changes in the body weight and treatment related change like respiration rate and heart rate.  Persistent treatment related changes were observed in behavioural signs viz apathy, reduced locomotor behavior but regained after 24 hrs. Consequently, 2000mg/Kg of plant extract found safe with less toxic effect.it test dose of 2000mg/Kg was used.
  • 19. SUB ACUTE TOXICITY STUDY The methanolic extract of Plectranthus amboinicus (Lour) Spreng at dose of 200,400 mg/kg orally for every 24 hr for 28 days did not produce any mortality in tested animals. No sign of observable toxicity was detected during the experimental period. All the tested haematological parameters such as hemoglobin, R.B.C, Platelet count, Reticulocyte count, Mean corpuscular volume, mean corpuscular hemoglobin concentration, Percent of Neutrophils, Lymphocytes and Monocytes, Packed cell volume and mean corpuscular hemoglobin were within normal. W.B.C count was slightly increased in group-II and decreased in Group-III than the Group-I. Biochemical parameters such as serum bilirubin, Serum glutamic oxaloacetic Transaminase, Serum Glutamic pyruvic Transaminase, Serum alkaline phosphatase, Serum total proteins, serum total albumin, serum total globulin and serum cholesterol and serum triglyceride were within the normal.
