Each component interacts differently with adsorbent
Separation of the components

6. TANDEM MASS SPECTROMETRY
MS/MS
MS1:
precursor ions are formed
separate by m/z ratio
Daughter ions are created by fragmentation
MS2:
Ions are separated and detected
Proteins are digested to produce a complex mixture of peptides, which are separated by HPLC before analysis by MS. The overall process consists of a number of steps, specifically the ionization of the peptides, acquisition of a full spectrum (survey scan) and selection of specific precursor ions to be fragmented, fragmentation, and acquisition of MS/MS spectra (product-ion spectra). The data are processed to either quantify the different species and/or determine the peptide amino acid sequence through a database search.
