Biphenyl aldehyde 4 was prepared in 73% yield by Suzuki coupling of 4-trifluoromethylbenzeneboronic acid 2
and 4-bromobenzaldehyde 3. The conversion of 4 to diamine 5 was achieved in 84% yield by reductive
amination with N,N-diethylethylenediamine. Diamine 5 was used in the next step without further purification.

Thiouracil 8 was synthesized in 64% yield by the condensation of ethyl 2-oxocyclopentanecarboxylate 6 and
thiourea 7. Thiouracil 8 was alkylated with 4-fluorobenzyl chloride to give 9 in 90% yield, which reacted with tert-
butyl bromoacetate to furnish adduct 10 in 87% yield. Cleavage of tert-butyl ester was achieved by treating with
trifluoroacetic acid, and product 11 was obtained in 95% yield in >98% purity after crystallization from EtOH.
Coupling of diamine 5 and carboxylic acid 11 in the presence of HBTU [94790-37-1], followed by crystallization
from isopropyl acetate gave Darapladib 1 in 60% yield and >99% purity.
Darapladib 1 exists in 1HNMR as a mixture of rotamers in about 1.2:1 ratio; and that ratio was 1.9:1 in literature.
1H-NMR and ESI-MS are consistent to its structure. The melting point of 125-126oC is identical to the report of
125oC.
