2. INTRODUCTION
• Human blood comprises of three components:
• Erythrocytes or Red Blood Cells,
• Leucocytes or White Blood Cells, and
• Platelets or Thrombocytes suspended in a fluid called plasma.-
• Packed cell volume test also known as Hematocrit test, refers to the ratio
occupied by Red Blood Cells to the total volume occupied by all the blood
components or whole blood (Erythrocytes, Leucocytes and Platelets) in a
given blood sample.- Packed Cell Volume test measures how much of the
blood consist of cells.
3. AIM
• To diagnose certain blood disorders such as anaemia (decrease in
RBCs), polycythemia ( Increase in RBCs) etc.
• For monitoring the overall health of patients.
4. PRINCIPLE
• A given volume of whole blood sample with anticoagulant is
centrifuged in a high speed centrifuge machine for 3-5minutes.
• This process results in the separation of the whole blood sample,
the denser (Erythrocytes) at the bottom of the capillary tube, the
buffy coat in between and the Leucocytes and Platelets suspended
in the plasma at the topmost part of the capillary tube.
• The denser part, that is, the Erythrocytes is measured using a
microhematocrit reader and recorded in percentage.
6. Procedure-
• collection of small quantity of whole blood from patient's vein using
a syringe- Transfer blood sample in a sample bottle, then collect
from the bottle in a capillary tube
• Seal the bottom end of the capillary tube with a sealant- Spin blood
sample in the capillary tube in a high speed centrifuge machine for
3-5 minutes
• Measure the Red Blood Cells (Erythrocytes) using a
microhematocrit reader and then record results in percentage.
8. CONCLUSION-
• Packed cell volume (PCV) Normal Range; Male:
• 41% to 50%
• Female: 36% to 44%
• A low packed cell volume (PCV) implies that the patient has a low number of Red
Blood Cells and is suffering from anaemia, which may be probed further with
other tests to determine the underlying cause of anaemia.
• Increased packed cell volume (PCV) can generally mean that a person is
dehydrated and there is a higher number of Red Blood Cells production, and is
suffering from polycythemia.
