Enzymes in Major Diagnostic Use
Enzyme Disease
Acid phosphatase Prostate cancer
Alkaline phosphatase Liver and bone disease
Amylase Acute pancreatitis
Aspartateaminotransferase Liver and heart disease
Alanineaminotransferase Viral hepatitis
Creatininekinase Muscle disorders and myocardial
infarction
Lactate dehydrogenase Myocardial infarction
Lipase Acute pancreatitis
2
ACID PHOSPHATASE(ACP)
It is basically phospho-monoestarase
Stored in lysosome
Different forms found in different
organs
Their serum level used in prostate
cancer
An assay on the rates of activity of
these enzymes may be used to
acertain biological demand for
phosphate in soil. Rajesh kumar sahu 3
ALKALINE PHOSPHATASE
(AP)
It is a glycoprotein with subunit
masses ranging from 40-83 kDa
AP found in intestine,kidney,liver and
bone
AP found in bile cannalicular
membrane of hepatocytes of healthy
dogs and rats
Also sinusoidal surface of human
being
Rajesh kumar sahu 4
CONTINUE….
In man there are two genes coding for AP
INTESTINAL AP( IAP)
TISSUE UNSPECIFIC AP ( UAP)
Increase of serum UAP denotes cholestasis
In dogs under the influnce of excess gluco
corticoids it produces hyper glycosylated
form of IAP called as CAP
CAP present in the serum of hyperadrenal
corticism and prolonged ill ness.
Rajesh kumar sahu 5
CREATININE KINASE (CK)
It has four iso enzymes which are
dimers of two protomers with
individual masses of 40 kDa
CK activated by Mg+2
Serum CK serves as indicator of
cardiac infarction
Bruising, prolonged recumbency,
convulsion, shievering results in
incresed CK
Rajesh kumar sahu 6
ALANINE AMINO
TRANSFERASE
Incresed ALT activity is seen in
hepatocyte damage
Human and porcine ALT are relatively
stable at room temp but when frozen
60% activity loss
Rajesh kumar sahu 7
ASPARTATE AMINO
TRANSFERASE
AST has two iso-enzymes cytosolic
and mitochondrial which has masses
about 92kDa
Presence of AST denotes soft tissue
damage,
Rajesh kumar sahu 8
SORBITOL
DEHYDROGENASE
Also called iditol dehydrogenase
Hepatocytes have high SDH activity
and incresed plasma SDH is
consistent of hepatocytes damage in
many species.
Rajesh kumar sahu 9
Lactate dehydrogenase
It is tetramers of two protomers with
molecular weight 35kDa
Hamolysis appreciable increses the
LDH
Anticoagulant such as EDTA and
oxaltes directly inhibit LDH activity
Rajesh kumar sahu 10
Cholinterase (ChE)
Has acytocholine as substrate
Organo phosphate insectisides,nerve
gas are the inhibitors
Decresed ChE in acute
infection,pulmonary infection,muscular
dystrophy,renal disease,insecticide
intoxication
Rajesh kumar sahu 11
Alpha amylase
Ca +2 depended
Pancreatic dis-order indiction
Rajesh kumar sahu 12
γ-glutamyl transferase
Physiological function is speculated
with glutathione metabolism.
Cholestatic disorder results in
incresed serum the enzyme activity.
Also indicator of nephro-toxicity
Bile duct disease also increses the
enzyme conc
