Introduction: Sanger DNA sequencing remains the most straightforward, reliable method for determining the
precise genetic sequence of DNA fragments. However, it requires a fairly significant investment in time and
materials. Mutation scanning strategies seek to quickly and efficiently scan DNA samples from many individuals
for minor genetic variations to identify candidates to be subjected to full sequencing analysis. High resolution melt
analysis (HRM), a technology that is based on the analysis of the melting profile of PCR products through gradual
temperature increase, is suitable for mutation scanning to Identify candidates for Sequencing.
Aim: The aim of this work is to apply qPCR-HRM for mutation detection based on the melting behavior of double
stranded DNA.
Method: DNA from 25 individuals was extracted from whole blood using salting out method then resuspended
in a low-salt buffer TE (10 mM Tris, 1 mM EDTA) prior to PCR. Primer couple was designed to amplify the target
sequence using primer3 software then specificity checked with BLAST tool. Amplification was performed into
Bio-Rad CFX96 thermo-cycler. Precision melt analysis software was used for melting curves analysis. Samples with
atypical shape are considered candidate to be subjected to sequencing in order to identify genetic variation. ABI
3100 genetic analyzer (Applied biosystem) was used to determine the nucleic acid sequence of candidate samples.
Results :We detected with HRM analysis one sample with shape and Tm different from the other samples and
forming a cluster separately. To confirm whether this change is due to a DNA variation or eventual troubleshooting,
the sample was sequenced. A single nucleotide variation was detected confirming the previous result of HRM.
Conclusion : This work allowed: (i) the application of qPCR-HRM method for mutation detection. (ii) Highlighting
