47. Que peut on proposer?
On a montré que :
- La stabilité de la ferroquine est compatible avec les conditions internes à la vaccuole
digestive (H2O2)
- Dans ces conditions on observe la formation de l’ion HO°
FQ est en millimolaire et HO° en micromolaire
--> mais HO° réagit en 10 -9 secondes donc autour de son site de formation*.
FQ par sa lipophilie s’accumule au niveau des membranes où a lieu la polymérisation
de l’hème. Le mécanisme conduisant à la résistance ne peut avoir lieu.
Le mécanisme d’attaque des acides gras par HO°produit de la malonaldéhyde (et du 4 et 8
hydroxynonénal) qui réagit sur les NH2 en bout de chaîne pour former des ponts entre les
protéines.
Ces ponts sont détectables par anticorps spécifiques.
--> La mise en évidence de ces anticorps est notre prochain objectif.
*Aucun mécanisme contre les radicaux libres = pas de détoxification possible

pyrimethamine plus sulfadiazine or clindamycin are the drugs of choice for therapy
but some patients are intolerant.
toxicity effects = allergies (especially in AIDS patients)
